Facility offers both infrastructure and scientific- technical support for the achievement of microscopy confocal studies. We can study different samples: fixed, living, cell cultures and tissues both from animal and plant origin; as well as diverse biomaterials. Equipment: Confocal Laser Scanning Microscope (CLSM) LEICA TCS SP8 STED 3X fitted witth UV Laser 405 nm and White Light Laser freely tunable excitation from 470-670 nm (1 nm steps). AOTF (Acousto-Optical Tunable Filter) and AOBS (Acousto-Optical Beam Splitter) System. Scanning stage for motorized stage applications. La presencia de marcadores fluorescentes en las muestras permite la utilización de una amplia variedad de aproximaciones experimentales en células vivas y fijadas. El servicio proporciona el equipamiento necesario para la realización de estudios de microscopía de epifluorescencia, confocal y superresolución mediante FLIM, FCS, DLS, STED, TIRF y SMLM, así como las herramientas informáticas para el procesamiento y análisis de las imágenes digitales generadas. El personal del servicio ofrece asistencia y entrenamiento en la utilización de los equipos, diseños experimentales, aplicaciones y metodologías de procesamiento/análisis de imagen. El servicio mantiene y proporciona un stock de reactivos, anticuerpos secundarios y soportes de cultivo ampliamente utilizados en las aplicaciones de microscopía de fluorescencia. El Servicio está certificado por la norma ISO 9001:2015, y forma parte de la Red de Laboratorios de la Comunidad de Madrid y la Red Española de Microscopía Óptica Avanzada.

El Servicio de Microscopía Confocal y Electrónica de Transmisión (CTEM) tiene como finalidad satisfacer la creciente demanda de los investigadores del Centro en este ámbito. El Servicio pretende organizar, gestionar, optimizar y fomentar el uso de los grandes equipos de microscopía disponibles en el Instituto. También facilita la preparación estándar de las muestras antes de su observación. La microscopía óptica es una de las herramientas de investigación fundamentales en el IBFG. Hoy en día, con el desarrollo y las aplicaciones de las proteínas fluorescentes, es posible el diseño de experimentos complejos que permiten estudiar la dinámica de los procesos celulares en células vivas. Estos experimentos pueden ser realizados adquiriendo imágenes de las muestras que proporcionan información multidimensional en x, y, z, longitud de onda y tiempo. El servicio de microscopía tiene una dotación de equipos que permiten trabajar con una amplia gama de técnicas de microscopía óptica, desde la adquisición de imágenes de campo claro y fluorescencia convencional, hasta las microscopías confocales espectral y “spinning disk”. El servicio también tiene el equipamiento informático con los programas necesarios para el procesamiento de las imágenes adquiridas en los experimentos.

El trabajo realizado por el servicio de Microscopía del IBGM, puede resumirse en dos aspectos.Por una parte, el destinado al uso del Microscopio Confocal Leica TCS SP5 X, y por otra, aquel en el que se realizan las labores de mantenimiento y gestión de los equipos de microscopía que dispone el Instituto. Las labores realizadas con el Microscopio Confocal Leica SP5, van desde la preparación de los settings adecuados para cada muestra, la adquisición de imágenes confocales de las muestras de los usuarios del Servicio y ayuda sobre los programas de tratamiento de imágenes. El trabajo derivado del mantenimiento y gestión de los equipos, consiste en dar una guía de uso de los mismos a los nuevos usuarios del Servicio, prestar ayuda cuando éstos encuentren algún tipo de problema, y chequear el buen funcionamiento de los equipos (microscopios de Fluorescencia Nikon).

El Servicio de Microscopía Confocal y Citometría del IBVF presta sus servicios tanto a los miembros del propio Instituto como a interesados externos, ya sean pertenecientes a organismos públicos como a entidades privadas. El Servicio incluye el encendido y puesta en marcha del equipo y la asistencia técnica por el personal responsable del servicio. El Servicio cuenta con un Citómetro Separador BD Influx, un Microscopio de Fluorescencia Leica DM6000 y un Microscopio de Fluorescencia Láser Confocal Leica SP2. Nuestros usuarios son principalmente del campo de la biología y biomedicina, aunque de forma ocasional, se han realizado estudios de materiales en microscopía confocal. Las muestras que usualmente analizamos son cultivos in vivo de cianobacterias o microalgas, tejidos vegetales (hojas, raíces y tallos) o plántulas enteras, cortes de tejidos vegetales y animales y superficie de materiales.

El Servicio de Microscopía Óptica Avanzada (SMOA) es un servicio central de apoyo a la investigación del Centro de Biología Molecular Severo Ochoa. El SMOA dispone de sistemas avanzados de microscopía óptica, así como equipamiento informático especializado en la visualización y el análisis de imágenes. El equipo técnico del SMOA gestiona y mantiene los equipos de microscopía e informáticos, brinda soporte científico-técnico y asesora y forma (mediante seminarios y sesiones teórico-prácticas) a los investigadores en las técnicas de microscopía óptica. Asimismo, el personal del SMOA almacena y distribuye reactivos relacionados con estas técnicas y trabaja para mantener el servicio actualizado, incorporando y configurando nuevos sistemas. El SMOA pertenece a la Red Española de Microscopía Óptica Avanzada (REMOA), a la Red de Laboratorios e Infraestructuras (RedLab) y a la Plataforma de Microscopía para Biociencias y mantiene una acreditación de calidad y control ISO 9001:2015 (AENOR).

We have six confocal laser scanning microscope (CLSM) systems that allow for increased optical resolution compared to widefield microscopes, both in XY and Z axes. These systems also support photomanipulation applications (FRAP, photoactivation, or photoconversion), FLIM, 3D image reconstruction, and spectral characterization and separation of multiple fluorophores using up to six laser lines, a white laser, and spectral detectors. Five of our confocal systems are equipped with incubation chambers with temperature and CO¿ control for long-term in vivo studies. We also offer a spinning disk confocal system that maintains confocality at high acquisition speeds. It includes two cameras for simultaneous acquisitions and a SoRa disk to enhance resolution. La unidad de microscopía e histología es un servicio de investigación que ofrece la posibilidad de preparar muestras y observarlas por microscopía óptica y confocal. La unidad provee acceso al siguiente equipamiento: Estación automática de deshidratación e inclusión de muestras en parafina, dos criostatos, un vibratomo, un micrótomo, cuatro microscopios de luz transmitida y epifluorescencia, dos de ellos invertidos y todos con microfotografía digital, un microscopio estereológico, un microscopio confocal, y un microscopio spinning disk. También se dispone de una workstation de última generación con diferentes softwares de análisis de imágenes: ImageJ/Fiji, Cell Profiler, LAS X y ZEN Blue. La unidad de histología y microscopia proporciona asistencia técnica desde el diseño de experimentos hasta el análisis de los resultados obtenidos. Su función es proveer un servicio de calidad en técnicas histológicas y microscopia óptica.

El laboratorio de Corrosión del CENIM/CSIC desarrolla su actividad investigadora en dos vertientes diferentes íntimamente inter-relacionadas. En la primera de ellas, se realizan estudios básicos encaminados a entender los mecanismos que conducen al deterioro de los metales en ambientes agresivos, así como el desarrollo de nuevos materiales y métodos de protección frente a la corrosión. Se trata por tanto, de una labor eminentemente científica. No obstante, toda la experiencia del grupo y el equipamiento también se pone a disposición de las empresas para aportar soluciones a los problemas de corrosión que aparecen en los materiales durante su vida en servicio.

Spectral Confocal Microscopy allows the study of samples with fluorescent labeling, making optical sections thereof. About the point sample is excited by a laser. The emission wavelength of that sample is higher than excitation, and is the latter when passing through a small aperture (pinhole) allows detection of a single focal plane. This advance in the field of microscopy enables us to study the three-dimensional samples, including its interior, in particular materials and allows imaging of the surface by reflection. The system also allows laser plots of surface microtopography. - Spectral Confocal Microscope Leica TCS SPE, with 3 lines Laser 488nm, 532 nm and 635 nm fully automated. Has a program MetaMorph image analysis available to the user. Confocal microscopy sessions may have technical assistance or not have it. Sessions with technical assistance: Lab technicians try to adjust the schedule and needs of the various working groups. This has been made sessions and different time durations to allow greater flexibility and adaptation to user needs with the lab. The available technical assistance sessions have the following times: There are four sessions: 8: 30-12: 00 // 9: 30-13: 00 // 13: 00-14: 30 // 14: 30 to 16:00 For more effective, except in special cases operation, please avoid booking more than 2 sessions per week. To schedule the job to do, is to get on initial telephone contact with the laboratory; depending on the timing of the users and the need for new work a series of initial sessions will be scheduled. The number and frequency of these will take place trying to complete the work of each user without accumulation of equal work sessions or one person takes place, in order to avoid too long waiting time for users. The sessions are to be provided well in advance to avoid long waiting lists and agglomerations in certain periods of greatest assistance due to foreign visitors, conferences or routine checks. Due to the long-term programming that takes place in some cases, it may be the fact of having to change and / or cancel any already scheduled session. In these cases we will try, where possible, to minimize impacts users trying to be fair with them. The reservation can be canceled at any time by notifying the technical manager of laboratory time to change shifts to another user. If cancellations consecutive repetition without giving notice, ie arriving at the day of the appointment, and consecutively the person concerned does not show, you can choose to collect the session although it has not enjoyed. Sessions WITHOUT technical assistance: NO technical assistance sessions are: 8:10 to 11: 30 // 11: 30-14: 30 // 14: 30 16:50 For more efficient operation, it should not exceed 6 hours of use per week per person. These users communicate technical staff incidents that may have occurred during handling equipment, in particular those that may require repair or maintenance. This will avoid inconvenience to users who want to use the computer in the next session. Routines and more delicate operations (the calibrated laser alignments and changes) MUST be done by lab technicians). El Servicio de Microscopía Óptica y Confocal del IIBm proporciona a sus usuarios una variedad de técnicas en microscopia acorde con las investigaciones que se realizan en el centro. Nuestros objetivos son dar soporte técnico tanto en la adquisición como en el análisis de las imágenes, formar a los usuarios en el uso de los equipos, participar en el desarrollo de nuevas técnicas y en la optimización de las ya establecidas para proporcionar los mejores resultados. Trabajamos bajo la norma de gestion de la calidad (UNE-EN ISO 9001:2000) y estamos certificados con el nº de registro ER-1326/2009. Nuestros equipos están disponibles para los usuarios del IIBm así como para investigadores de otras instituciones públicas o privadas. La misión del Servicio es prestar soporte y asesoramiento a los organismos públicos y privados que así lo soliciten. El servicio consta de tres divisiones: Caracterización de Superficies y recubrimientos: cuenta con un perfilómetro 3D óptico, un nano-indentador y un microscopio de sonda local, que, permiten realizar topografías de superficies, desde los milímetros a los nanómetros, así como estudios de propiedades elásticas y otras propiedades físicas en el rango de la nanoescala. Análisis Elemental: permite la determinación de la composición porcentual de carbono, hidrógeno, nitrógeno y azufre, de forma simultánea, de una muestra o compuesto en estado sólido o líquido. Análisis Térmico: estudia los cambios producidos en alguna propiedad del material en función de la temperatura. Cuenta con equipos de calorimetría diferencial de barrido (determinación de absorciones y emisiones de calor vs temperatura), y termogravimetría (determinación la variación de masa vs temperatura

Several methods allow the construction of 3D profiles of surfaces and are used depending on the type of sample to be studied. The equipment has confocal and interferometry modes. In the confocal microscopy technique, a diaphragm eliminates out-of-focus light, and the 3D map is constructed with the images obtained at various heights.
