FASP (Filter Aided Sample Preparation. See Wisniewski JR et al, 2009).
A method for the treatment of proteins which are obtained with high concentrations of SDS (insoluble, membrane proteins) or other substances interfering with trypsin digestion.
Between 25-150 µg of protein are loaded into an Amicon centrifugal filter (10-kDa cutoff). The sample in the filter is washed with 8 M Urea buffer followed by centrifugation. Proteins retained in the filter are alkylated and digested in situ. Tryptic peptides are recovered by addition of 3×100 µl of buffer and centrifugation.
