Description:
DNA fragmentation is a key step for genomic library construction and its subsequent application to high throughput sequencing protocols, among other applications (see details on the applications section). Among the different methods available (physical, chemical and enzymatic shearing), our service offers fragmentation by sonication, a process which consists of hydrodynamic shearing of DNA samples through short periods of controlled sonication. This process is highly efficient in comparison to other methods, reaching high levels of sensitivity. It is reproducible and not influenced by technical manipulation, laboratory conditions and change of equipment used.
Equipment available:
Qsonica Sonicator Q800R3 (https://www.sonicator.com/products/q800r-sonicator). DNA and chromatin shearing system in liquid. This equipment allows fragmentation of DNA within a range of sizes considerably wide (from 150 bp to 3 Kb). Fragment size needs to be optimized and will depend on the methodological goal. The system allows simultaneous multiplexing of up to 18 samples. The volumen required per sample ranges between 50 µl and 1 mL. Standard comercial tube sizes for molecular lab experiments are used (0.3 mL PCR tubes, 0.5 mL PCR PCR tubes and 1,5 mL poliestirene Eppendorf tubes).
